The GRT Hub provides several services for single-cell profiling. We currently have four options for single-cell profiling: Fluidigm C1 single-cell prep, 10x Genomics Chromium single-cell assays, Mission Bio Tapestri, and Parse Biosciences single-cell assays. Instruments for the Fluidigm C1, 10x Genomics Chromium, and Mission Bio Tapestri workflows are available for self-operation or full service.

Single-cell profiling assays include targeted DNA, gene expression, ATAC, immune (VDJ), and protein profiling.

Sample Submission Guidelines

  • In general, for single-cell assay, you should generate a clean and viable single-cell/nuclei suspension.
  • This suspension should be free of debris and ambient RNA, and there should not be any cell/nuclei clumps. The viability of the cell or nuclei should be as high as possible. Inputting in lower-quality cells or nuclei will result in poor-quality data.
  • Final input into each assay varies. Please see below for the requirements for each platform.

Parse Biosciences Single-cell Assay

  • Fix and freeze your single-cell suspension according to the latest Parse Biosciences fixation protocol. This requires special fixation kits from Parse Biosciences. There are two different kits, one for cells and another for nuclei

10x Genomics Chromium

  • For 3’ and 5’ assay, please prepare a single-cell suspension that is at about 1,000 cells/uL. For ATAC and Multiome assay, please prepare a single-cell suspension that is at about 5,000 cells/uL. For the FLEX assay, you should fix and freeze your single-cell suspension according to the latest 10x Genomics FLEX fixation protocol. This requires a special fixation kit from 10x Genomics.

Mission Bio Tapestri

  • You should prepare a single-cell suspension that is about 3,000 cells/uL.

Fluidigm C1

  • You should prepare a single-cell suspension that is about 2,000 cells/uL.
